Simple identification method to distinguish Bacillus pumilus from B. zhangzhouensis (2024)

Abstract

Bacillus pumilus and B. zhangzhouensis are two major bacterial strains isolated from Doenjang, a traditional Korean fermented food. However, identification of these species is challenging because of their similarity in the 16s rKNA region. Instead of relying on 16s rRNA sequencing, this study explored the possibility of distinguishing between these two strains based on the morphology of colonies grown on solid media. Four types of media (BHI, BL, PC, and TS) were used to observe the morphological characteristics of colonies. The results showed that it was easy to differentiate B. pumilus and B. zhangzhouensis in BL medium. Therefore, a simple and cost-effective way to analyze these strains was proposed using one-way PCR sequencing of the 16s rRNA region and evaluation of their growth patterns on BL solid medium. In this method, one-way PCR sequencing was useful for distinguishing B. pumilus and B. zhangzhouensis from other Bacillus strains, and the growth pattern in BL medium distinguished B. pumilus from B. zhangzhouensis.
